The interaction of cell surface TLR receptors with LPG, which signals cellular responses during\u00a0<em>Trypanosma brucei<\/em>\u00a0infection, was systematically investigated.<\/h5>\n<p>The cytokine expression profile in neutrophils after exposure to\u00a0<em>T. brucei<\/em>\u00a0LPG, and the involvement of TLR2, TLR4, p38 MAP kinase, and NF-\u03baB in NET formation were studied with molecular immunological approaches including quantitative PCR, western blotting and immunofluorescence.<\/p>\n<ol>\n<li><em> brucei<\/em>-derived LPG induced phosphorylation of p38 MAP kinase and NF-\u03baB, thereby stimulating neutrophil secretion of IL-1\u03b2, IL-8, and TNF-\u03b1. The blockade of Toll-like receptor 2\/4 and specific inhibitors of MyD88, p38 MAP kinase, and NF-\u03baB decreased cytokine release and the phosphorylation of both kinases.
