This work was aimed at increasing epidemiological knowledge of\u00a0<em>Rickettsia conorii<\/em>\u00a0and\u00a0<em>Rickettsia slovaca<\/em>\u00a0in a geographical area in Castilla-La Mancha (Spain) lacking prior data, through a seroprevalence study in potential wild rodent reservoirs.<\/p>\n<p>A serological survey collected 118 rodent sera from field (79) and peri-urban (39) areas through convenience sampling between April and September 2019 in Montes de Toledo (56.7%\u00a0<em>Mus spretus<\/em>, 24.6%\u00a0<em>Mus domesticus<\/em>, 10.1%\u00a0<em>Apodemus sylvaticus<\/em>, and 8.5%\u00a0<em>Rattus norvegicus<\/em>). Sera were tested for antibodies to\u00a0<em>R. conorii<\/em>\u00a0and\u00a0<em>R. slovaca<\/em>\u00a0through indirect immunofluorescence assay (IFA).<\/p>\n<p>Antibodies to\u00a0<em>R. conorii<\/em>\u00a0were detected in 62\/118 rodents (52.5%). The highest prevalence was observed in\u00a0<em>R. norvegicus<\/em>\u00a0(90%) and\u00a0<em>M. spretus<\/em>\u00a0(62.7%). In field areas and peri-urban areas, the prevalence was 53.2% vs 51.3%, respectively (\u03c7<sup>2<\/sup>=0.04; p=0.847). Antibodies to\u00a0<em>R. slovaca<\/em>\u00a0were found in 22\/118 (18.6%), primarily in\u00a0<em>R. norvegicus<\/em>\u00a0(90%) and\u00a0<em>M. spretus<\/em>\u00a0(17.9%), and did not show differences between field and peri-urban environments (15.2% vs 25.6%; \u03c7<sup>2<\/sup>=1.88; p=0.17).
