Coinfection with\u00a0<em>Cryptosporidium<\/em>\u00a0spp. may worsen clinical outcomes but has been understudied in Iraq. This study assessed the prevalence of\u00a0<em>H. pylori<\/em>\u2013<em>Cryptosporidium<\/em>\u00a0coinfection and performed phylogenetic analysis of local isolates.<\/p>\n<p>A cross-sectional study was conducted in 420 gastrointestinal patients in Thi Qar, Iraq.\u00a0<em>Cryptosporidium<\/em>\u00a0was detected through modified Ziehl-Neelsen staining and nested PCR (SSU rRNA).\u00a0<em>H. pylori<\/em>\u00a0was identified through stool antigen testing and PCR (16S rRNA). Nine\u00a0<em>H. pylori<\/em>\u00a0isolates (479 bp) and coinfecting\u00a0<em>Cryptosporidium<\/em>\u00a0isolates were sequenced. SNPs were analyzed, and a UPGMA tree (1,000 bootstraps) Results:<\/p>\n<p>The prevalence was 51.4% for\u00a0<em>H. pylori<\/em>, 70.5% for\u00a0<em>Cryptosporidium<\/em>, and 37.6% for coinfection. The sequence identity with respect to the reference strain was 99.10%\u201399.74%. The 16S rRNA alignment revealed nine polymorphic sites with a conserved G\u2192A transition at position 200, and seven haplotypes. A putative novel A\u2192T transversion at position 146 of the\u00a0<em>Cryptosporidium<\/em>\u00a0SSU rRNA gene was detected in a coinfected isolate (IQN.CO1). GenBank accessions:\u00a0<em>H. pylori<\/em>\u00a0PZ245395\u2013PZ245403;\u00a0<em>Cryptosporidium<\/em>\u00a0PZ245405\u2013PZ245413 (SUB16104315).<\/p>\n<p>High coinfection prevalence suggested epidemiological overlap. Both\u00a0<em>C. hominis<\/em>\u00a0and\u00a0<em>C. parvum<\/em>\u00a0were identified, thus indicating diverse transmission routes.
