The purpose of this study was to map the bacterium flora and identify the main pathogens in ticks in grazing areas of Yunnan province.<\/p>\n<p>The V3-V4 region of 16S rRNA amplifier sequencing was used to analyze the tick-borne pathogens in\u00a0<em>Rh. Microplus<\/em>. A prevalence survey on\u00a0<em>B. burgdorferi s.l.<\/em>,\u00a0<em>B. miyamotoi<\/em>,\u00a0<em>E. chaffeensis<\/em>,\u00a0<em>A. phagocytophilum<\/em>,\u00a0<em>Coxiella burnetiid<\/em>, and the spotted fever group\u00a0<em>rickettsiae<\/em>\u00a0was carried out using PCR. Phylogenetic analysis was used to identify and cluster the main prevalent microbe amplicons.<\/p>\n<p>One hundred five genera and 117 species were detected in 50 ticks. <em>Rickettsia<\/em>,\u00a0<em>Anaplasma<\/em>,\u00a0<em>Borrelia<\/em>,\u00a0<em>Coxiella<\/em>, and\u00a0<em>Ehrlichia<\/em>\u00a0were identified in tick samples by high-throughput sequencing at the genus level. Pathogen prevalence testing showed that the ticks were positive for\u00a0<em>B. burgdorferi s.l.<\/em>\u00a0(9\/50 [18%]),\u00a0<em>A. phagocytophilum<\/em>\u00a0(1\/50 [2%]), and the spotted fever group\u00a0<em>rickettsiae<\/em>\u00a0(35\/50 [70%]), which were closely related to\u00a0<em>Candidatus R. jiangxinensis<\/em>.<\/p>\n<p>The spotted fever group\u00a0<em>rickettsiae<\/em>\u00a0was the dominant pathogens in Xishuangbanna. Dual co-infection (1\/50 [14%]) with\u00a0<em>Candidatus R. jiangxinensis<\/em>\u00a0and\u00a0<em>Borrelia garinii<\/em>\u00a0was demonstrated. V3-V4 high-throughput sequencing of the 16S rRNA gene was not sensitive to identify species for some bacteria, so more accurate and comprehensive analysis is necessary.<\/p>\n<p><a href=\"https:\/\/www.scienceopen.com\/hosted-document?doi=10.15212\/ZOONOSES-2023-0040\">https:\/\/www.scienceopen.com\/hosted-document?doi=10.15212\/ZOONOSES-2023-0040<\/a><\/p>\n<p><em>Zoonoses<\/em> is fully open access journal for research scientists, physicians, veterinarians, and public health professionals working on diverse disciplinaries of zoonotic diseases.
